I'm developing a new suspension formulation, and one of the issues we have is the bioavailability of the active ingredient, which is poor because of limited solubility. We're exploring pharmaceutical wet milling as a way to reduce particle size to <1 micron; however, I worry that the heat generated during this process will decompose the active ingredient before milling is complete. Has anyone here found a way to control the temperature during high-energy milling? I'd love to hear your experiences or recommendations regarding optimization of process parameters in order to maintain a lower product temperature.
